Targeted Discovery for miRNA

nCounter® miRNA Expression Assays have been carefully curated to ensure the relevant content and appropriate controls to achieve excellent research results. NanoString utilizes proprietary metrics such as observed read ratios and expression analytics to screen potential content before inclusion in the panel.

These methods help ensure that miRNA panel content is heavily weighted toward biologically significant miRNAs. In addition to this data-driven selection process, NanoString also performs literature reviews to ensure that the most relevant miRNAs are included in our panels.

Controls Ensure Robust Performance

Each nCounter® miRNA panel contains a large number of control probes to ensure robust performance. For the newest panel versions, Human v3 miRNA, Mouse v1.5 miRNA, and Rat v1.5 miRNA, NanoString's miRNA panel controls have been updated to include probes for both hybridization and ligation based on sequences generated by the ERCC (External RNA Controls Consortium).

miRNA Profiling with Excellent Accuracy

nCounter miRNA Expression Assays enable highly multiplexed, direct digital detection and counting of miRNAs in a single reaction without amplification. NanoString offers convenient, fixed-content panels for analysis of miRNA transcriptomes of Human, Mouse, and Rat. nCounter's unbiased and simple workflow enables these panels to generate miRNA expression profiles with great sensitivity and reproducibility. Custom offerings are also available enabling the analysis of focused sub-sets of miRNAs (nCounter Custom à la carte miRNA CodeSets).

Reproducibility

To demonstrate the reproducibility of data generated with the nCounter Human v3 miRNA Expression Kit, we prepared an experiment, using 100 ng total RNA per replicate sample preparation. Counts for individual probes in technical replicates within a cartridge were highly correlated between sample preparations with R² > 0.99.

FFPE Performance

Similarly, a comparison of nCounter data generated after preparation of total RNA from FFPE tissue and matched frozen (non-FFPE) tissue also showed highly significant concordance, with R² > 0.95.
