The nCounter Analysis is based on gene-specific probe pairs that are hybridized to the sample in solution. The CodeSet is the heart of our system and consists of the probe pairs that will hybridize with your genes of interest. Each gene is targeted with two consecutive probes, the Capture Probe and the Reporter Probe. The Capture Probe is used to immobilize the target complex for data collection. The Reporter Probe carries the barcoded fluorescent signal that identifies the target. We currently provide CodeSets that can assay up to 550 genes. All the genes are profiled in a single reaction. The high sensitivity of the assay enables you to use smaller amounts of starting material. The assay works with a wide range of sample types including total RNA, cell lysate, FFPE samples and whole-blood lysate. All the reagents and consumables to run the assay are provided by NanoString in the nCounter Master Kit.

nCounter Gene Expression Panels
nCounter Gene Expression Panels are off-the-shelf preassembled CodeSets. CodeSet Panels include all of the consumables to perform the assay. Gene Lists for the nCounter Gene Expression Panels were created by cross referencing several of the public databases.

nCounter Virtual Gene Sets
nCounter Virtual Gene Sets are comprehensive lists of genes for the study of genetic pathways, disease states and biological processes. Genes included in the nCounter Virtual Gene Sets are selected based on empirical data obtained from public databases and tools such as Ingenuity Pathway Analysis® from Ingenuity® Systems, Inc. CodeSets built from the Virtual Gene Sets are made-to-order and rapidly shipped. NanoString provides all of the reagents and consumables to run the assay in the nCounter Master Kit, which must be ordered separately.
nCounter Custom CodeSet
NanoString enables you to assay the specific genes you wish to study. We have developed a CodeSet design and manufacturing process that will meet all of your needs. The process starts with the probe design services provided by our Bioinformatics Team. You simply provide us with the list of NCBI Reference Sequence (RefSeq) IDs you want to target and we will design the best probes for your custom multiplex assay. Once the design is approved by you, our Manufacturing Group will assemble your CodeSet along with all of the reagents and consumables needed to run the number of samples you have specified for your project. All custom CodeSets undergo extensive quality-control testing before they are sent to you to ensure your results meet our very high standards.
