BENCHMARKING TO GOLD STANDARD TECHNIQUES
Combined RNA and protein profiling reveals potential post-transcriptional regulation for a subset of targets and protein expression data that is highly correlated with gold standard flow cytometry.
PBMCs were stimulated with PMA and RNA and protein expression was profiled using the nCounter Vantage 3D RNA:Protein Immune Cell Profiling Assay (input of 50,000 PBMCs). Additionally, a subset of protein targets were analyzed by flow cytometry and data from both platforms showed high correlation.
CHARACTERIZING MACROPHAGE DIFFERENTIATION AND POLARIZATION

A THP-1 monocytic cell line was induced to differentiate into a macrophage phenotype with PMA. Cells were then polarized to an M1 or M2 phenotype by 24h culture using IFNγ + LPS, or IL4, respectively. Gene and protein expression was characterized at each stage of differentiation using the nCounter Vantage 3D RNA:Protein Immune Cell Profiling Assay.
